試劑盒組成

● 1或3(以規格而定)玻管Lightening-Link™ Mix 混合物。 

● 1管LL-X（X為(wei) 標記物，如PE或HRP等）修飾試劑。 

● 1管LL-X萃滅試劑。

操作步驟

1）在每10ul抗體(ti) 樣品中加入1ul的LL-Modifier修飾試劑，輕輕混勻

2）打開Ligtening-Link混合物管蓋，吸取步驟1已混好的抗體(ti) 樣品至管內(nei) 凍幹混合物中，用吸液器輕輕吸打1-2次,使粉末*溶解

3）蓋上玻璃瓶蓋，室溫(20-25°C)避光孵育3小時,或過夜

4）孵育3小時後，按每10ul抗體(ti) 的量加入1ul LL-淬滅試劑，靜置30分鍾後即可使用

 *不同的標記物，操作步驟略有不同。（操作步驟供參考，以英文原版說明書(shu) 為(wei) 準

標記結合物的保存

對於(yu) 任何新結合物，推薦保持於(yu) 4°C條件下。如需長期保存，可添加保護劑

緩衝(chong) 液及添加劑兼容性：

A. 與(yu) 無胺基緩衝(chong) 液兼容，包括MES,MOPS,HEPES（濃度10-50mM, pH範圍6.5-8.5）,LL-Modifier修飾劑提供結合反應的條件

B. 常見的鹽（如氯化鈉）、螯合劑（EDTA）、糖對偶聯反應無影響

C. 疊氮鈉（0.02%-0.1%）對結合反應有輕微影響，疊氮鈉不可用於(yu) HRP標記，其為(wei) 後者的抑製劑

D. 甘油(up to 50%)對結合效率無影響

E. 若無胺基的緩衝(chong) 液濃度相對較高，pH值在6.6-8.5範圍外，則需要加入更多的LL-midifier修飾劑（/10ul抗體(ti) ），以此來調整pH值。的pH範圍為(wei) 7.3-7.6左右

F. 避免使用含親(qin) 核劑的緩衝(chong) 液，避免還原劑如DTT和2-巰基乙醇，乙醇胺等

G. 濃度達到或稍低於(yu) 20mM的Tris對結合效率有輕微的影響
